General description

Microtubules are constituent parts of the mitotic apparatus, cilia, flagella, and elements of the cytoskeleton. They consist principally of 2 soluble proteins, alpha- and beta-tubulin, each about 55 kDa in size.
Antibodies against beta Tubulin are useful as loading controls for Western Blotting. However it should be noted that levels of beta Tubulin may not be stable in certain cells. For example, expression of tubulin in adipose tissue is very low (Spiegelman and Farmer, Cell, 1982, 29(1):53-60) and therefore beta Tubulin should not be used as loading control for these tissues.

Specificity

Predicted to have broad species cross-reactivity based on sequence homology.
This antibody recognizes β-Tubulin isotypes I, II, III, IVa, and IVb of ~50 kDa. No cross-reactivity with alpha or gamma tubulin.

Immunogen

Epitope: The epitope has been mapped to amino acids 412-430
Purified bovine brain tubulin. The epitope recognized by this antibody has been mapped to amino acids 412-430.

This Anti-β-Tubulin Antibody, clone AA2 is validated for use in WB for the detection of β-Tubulin.

Quality

Routinely evaluated by western blot on RIPA lysates from 3T3/A31, PC-12, Jurkat, or A431 cells.

Western Blot Analysis:
0.1-1.0 µg/mL of this lot detected β-Tubulin in RIPA lysates from 3T3/A31, PC-12, Jurkat, and A431 cells.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.

Handling Recommendations:
Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20ºC. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20ºC may cause glycerol-containing solutions to become frozen during storage

Control
Included Positive Antigen Control: Catalog # 12-301, non-stimulated A431 lysate. Add 2.5 µL of 2-mercaptoethanol/100 µL of lysate and boil for 5 minutes to reduce the preparation. Load 20 µg of reduced lysate per lane for minigels.
