Descrizione generale

α-Ketoglutarate (α-KG) is a key intermediate in the Tricarboxylic Acid (TCA) Cycle, formed by the oxidative decarboylation of isocitrate by isocitrate dehydrogenase. The synthesis of α-KG from glutamate by glutamate dehydrogenase is a key TCA cycle anaplerotic reaction. α-KG is an important nitrogen transporter. This α-Ketoglutarate Assay Kit provides a simple, sensitive and rapid means for quantifying α-KG in a variety of samples.

Applicazioni

α-Ketoglutarate Assay Kit has been used to determine the activity of α-ketoglutarate.

Compatibilità

Suitable for the measurement of a-Ketoglutarate in a variety of biological samples including cell and tissue lysates

Principio

α-KG concentration is determined by a coupled enzyme assay, which results in a colorimetric (570 nm)/ fluorometric (λex = 535/λem = 587 nm) product, proportional to the α-KG present. Typical sensitivities of detection for this kit are 0.01 to 10 nmoles.

Isocitrate dehydrogenase 1 (IDH1) mutations are common in gliomas, acute myeloid leukemia, and chondrosarcoma. The mutation 'hotspot' is a single arginine residue, R132. The R132H mutant of IDH1 produces the 2-hydroxyglutarate (2-HG) carcinogen from α-ketoglutarate (α-KG). The reduction of α-KG

Articoli

Sigma-Aldrich presents an article about how proliferatively active cells require both a source of carbon and of nitrogen for the synthesis of macromolecules. Although a large proportion of tumor cells utilize aerobic glycolysis and shunt metabolites away from mitochondrial oxidative phosphorylation, many tumor cells exhibit increased mitochondrial activity.

Protocolli

We describe here a rapid and sensitive method to separate and measure D-2-OHG and L-2-OHG enantiomers using high-resolution mass spectrometry (HRMS) detection.
