Specificity

Clone 1M17 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects HSPG2 (Perlecan). It targets an epitope within 25 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal region of human HSPG2 (Perlecan).

Application

Quality Control Testing

Evaluated by Western Blotting in Human umbilical vein endothelial cells (HUVEC) lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected HSPG2 in lysate from Human umbilical vein endothelial cells (HUVEC).

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected HSPG2 in lysates from SH-SY5Y and HeLa cells and Rat heart tissue.

Affinity Binding Assay: A representative lot of this antibody bound HSPG2 peptide with a KD of 1.1 x 10-9 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HSPG2 in HT-29 cells.

Immunohistochemistry (Paraffin) Analysis: 1:100 from a representative lot detected HSPG2 in Human kidney tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Basement membrane-specific heparan sulfate proteoglycan core protein (UniProt: P98160; also known as HSPG, Perlecan, PLC) is encoded by the HSPG2 gene (Gene ID: 3339) in human. HSPG2 is a large basement membrane heparan sulfate proteoglycan that is expressed in a wide array of tissues where it regulates diverse cellular processes including bone formation, inflammation, cardiovascular development, and angiogenesis. It is a major basement membrane component and colocalizes with elastin in blood vessels, which contributes to their visco-elastic properties in vasculogenesis during establishment of early vascular networks and tissue development. It is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to produce the mature protein that can be further proteolytically cleaved into Endorepellin (aa 3687-4391) and a LG3 peptide (aa 4197-4391). It is a modular multifunctional proteoglycan with five distinct domains and each domain has a specific function. Its protein core is divided into several unique structural regions, each imparting distinct biofunctional diversity. It is reported to interact with several other basement membrane components, including laminin, prolargin, and collagen type IV. It also displays a strong tendency to aggregate in dimers or stellate structures. Its N-terminal region contains the primary location for heparan sulfate side-chain attachment, and this region contains three areas comprising the signature Ser-Gly-Asp (SGD) sequence. Mutations in HSPG2 gene have been linked to Schwartz-Jampel syndrome, a rare autosomal recessive disorder that is characterized by permanent myotonia and skeletal dysplasia. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
