General description

Monoclonal Anti-Tensin 3 (mouse IgG1 isotype) is derived from the hybridoma TN-17 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide corresponding to an internal sequence of human Tensin 3. Tensins are multidomain proteins that are characterized with homologous C1, protein phosphatases (PTPases) , C2, Src homology 2 (SH2) domains and pTyr-binding (PTB) domains.
Tensin 3 (TNS3) is encoded by the gene mapped to human chromosome 7p12.3. TNS3 transcript is expressed at high level in thyroid and at moderate level in placenta. In addition, the protein is also expressed at minimal levels in brain, heart, lung, liver, pancreas, kidney, skeletal muscle, white blood cells and prostate. TNS3 is a member of tensin protein family.

Immunogen

synthetic peptide corresponding to an internal sequence of human Tensin 3, conjugated to KLH. The isotype is determined using a double diffusion immunoassay using Mouse Monoclonal Antibody Isotyping Reagents (Sigma ISO-2).

Application

Monoclonal Anti-Tensin 3 antibody produced in mouse has also been used in immunofluorescence.
Monoclonal Anti-Tensin 3 antibody produced in mouse has been used in western blotting.

Biochem/physiol Actions

Tensin 3 (TNS3) plays a vital role in stimulation of epidermal growth factor (EGF) signaling pathway. The encoded protein supports fundamental role of thyroid hormone in normal thyroid tissues. Decreased expression of the gene leads to development of human renal cell carcinoma (RCC).
Tensin 3 facilitates cell migration and tumorigenesis in cell lines from advanced lung cancer, breast cancer and melanoma. In these cell lines, The protein is phosphorylated by Src in its SH2 domain, leading to tumorigenesis and metastasis, implicating tensin 3 as an oncoprotein regulated by Src.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
