General description

Specificity

Clone 2B9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects PKC . It targets an epitope within 21 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human PKC .

Application

Quality Control Testing

Evaluated by Western Blotting in Rat brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PKC in Rat brain tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PKC in Human brain and Rat brain tissue lysates.

Affinity Binding Assay: A representative lot of this antibody bound PKC with a KD of 1.4 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PKC in human cerebellum tissue sections.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected PKC in human cerebellum tissue sections.

Target description

Protein kinase C gamma type (UniProt: P05129; also known as EC:2.7.11.13, PKC-gamma) is encoded by the PRKCG (also known as PKCG) gene (Gene ID: 5582) in human. Protein kinase C (PKC), a ubiquitous, phospholipid dependent serine/threonine kinase that is involved in signal transduction associated with cell proliferation, differentiation, and apoptosis. At least eleven closely related PKC isozymes have been reported that differ in their structure, biochemical properties, tissue distribution, subcellular localization, and substrate specificity. PKCγ is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine kinase that plays diverse roles in neuronal cells and eye tissues. It binds and phosphorylates GRIA4/GLUR4 glutamate receptor and regulates its function by increasing plasma membrane-associated GRIA4 expression. The catalytic domain of PKCγ contains an ATP-binding site. Its regulatory domain has a Ca2+ binding site. The amino terminal half of the regulatory domain contains two conserved regions, C1 and C2 that play a vital role in the regulation of enzyme activity. The catalytic domain is composed of highly conserved C3 and C4 regions. The C3 region contains the ATP-binding consensus sequence, whereas the C4 region is responsible for protein substrate binding. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
