Specificity

Clone 2D9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects JAK3 phosphorylated on tyrosine 980/981.

Immunogen

KLH-conjugated linear peptide corresponding to 10 amino acids surrounding phosphotyrosine 980/981 from human JAK3.

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected JAK3 in HepG2 cell lysate.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected phospho-JAK3 (Tyr 980/981) in HepG2 cells.

Affinity Binding Assay:: A representative lot of this antibody bound phospho-Jak3 (Tyr980/981) peptide with a KD of 9.2 x 10-7 in an affinity binding assay.

Flow Cytometry Analysis: 1 µg from a representative lot detected phospho-JAK3 (Tyr980/981) in one million NK-92 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Tyrosine-protein kinase JAK3 (UniProt: P52333; also known as EC:2.7.10.2, Janus kinase 3, JAK-3, Leukocyte janus kinase, L-JAK) is encoded by the JAK3 gene (Gene ID: 3718) in human. JAK/STAT signaling pathway plays an important role in cell proliferation, cell differentiation, cell migration, and cell death. It is the principal signaling mechanism for a variety of cytokines and growth factors. Constitutive activation or dysregulation of JAK/STAT signaling can result in inflammatory disease, erythrocytosis, gigantism, and leukemia. JAK3 is a non-receptor tyrosine kinase that mediates essential signaling events in both innate and adaptive immunity and plays a crucial role in hematopoiesis during T-cells development. It also mediates the secretion of IL-2, IL-4, and IL-9 that have been implicated in allergic inflammation. In the cytoplasm, it plays a pivotal role in signal transduction via its association with type I receptors sharing the common subunit gamma, which is a component of the IL-2 receptor and other interleukin receptors. Following ligand binding to cell surface receptors, it phosphorylates specific tyrosine residues on the cytoplasmic tails of the receptor, creating docking sites for STAT proteins. Subsequently, it phosphorylates STAT proteins following their recruitment to the receptor. Phosphorylated STATs then form homodimer or heterodimers and translocate to the nucleus to activate gene transcription. JAK3 possesses two phosphotransferase domains (aa 521-781 and 822-1111) and the second domain contains the catalytic region. JAK3 undergoes tyrosine phosphorylation on residues 980 and 981 in response to IL-2 and IL-4 and is dephosphorylated by protein tyrosine phosphatase non-receptor type 2 (PTPN2) that negatively regulates cytokine-mediated signaling. In humans, mutations in JAK3 gene have been associated with severe combined immunodeficiency disease. Three isoforms of JAK3 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
