General description

Protein inhibitor of activated STAT protein 3 (PIAS3) is an inhibitor of activated STAT3. The gene encodes for a 619-amino acid protein and has a molecular mass of 68 kDa. PIAS3 shares 55% identity with PIAS1 and PIASX proteins and 39% identity with PIASY. PIAS3 is widely expressed in various human tissues.

Biochem/physiol Actions

Protein inhibitor of activated STAT protein 3 (PIAS3) and other members of this family can act as small ubiquitin-like modifier (SUMO) E3 ligases and interfere with the transcription activity of many transcription factors. PIAS3 binds to microphthalmia-associated transcription factor (MITF), a key DNA-binding protein. It blocks MITF DNA-binding activity and co-expression of MITF. PIAS3 in NIH 3T3 cells inhibits MITF-driven transcription activity.
