General description

An immunoglobulin has two heavy chain and two light chain connected by disulfide bond. It mainly helps in immune defense. It is a glycoprotein. IgG is a major class of immunoglobulin. Rat has five immunoglobulin classes- IgM, IgG, IgA, IgD and IgGE. IgG has four subclasses- IgG1, IgG2a, IgG2b and IgG2c.
IgGs are glycoprotein antibodies that modulate several immune responses. Rat IgGs against target proteins are often used as primary antibodies in various research applications. Thus, secondary anti-rat IgGs conjugated to a detectable substrate are useful tools for the analysis of target proteins.
Anti-Rat IgG (whole molecule)-Peroxidase antibody binds all rat Igs and does not react with human IgG.

Application

Anti-Rat IgG (whole molecule)-Peroxidase antibody has been used as a secondary antibody in immunohistochemical applications at a dilution of 1:1500. The product has also been used for western blot analysis at 1:20,000. Furthermore, the antibody can be used for direct ELISA at 1:50,000.

HA-tagged proteins over expressed in S. cerevisiae were detected by western blot using HRP-conjugated rabbit anti-rat IgG as the secondary antibody.

Biochem/physiol Actions

Immunoglobulin (IgG) helps in opsonization, complement fixation and antibody dependent cell mediated cytotoxicity. IgG participates in hypersensitivity type II and type III.

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 0.05% MIT

Preparation Note

Adsorbed with human IgG to reduce background staining with human samples.
Prepared by two-step glutaraldehyde method.
