General description

Interferons are cytokines secreted by mammalian cells. They are broadly divided into type I– interferon alpha (IFN-α) and interferon beta (IFN-β), type II – interferon-gamma (IFN-γ), and type III – interferon lambda (IFN-λ) based on their structure, function, and receptor specificity. Recombinant αA interferon is a hydrophobic protein belonging to the IFN-α family of proteins.

Application
Interferon alpha has been used to evaluate its effect on VSV (vesicular stomatitis virus) infection in pancreatic ductal adenocarcinoma (PDA) cell lines and study the relationship between type I IFNsignaling, resistance to VSV infection, and the JAK/STAT (Janus kinase/signal transducers and activators of transcription) signaling pathways in PDA cell lines. It has also been used to study the inhibitory effect of Morbillivirus V Proteins on Interferon Signaling Pathways.
Biochem/physiol Actions
Recombinant, human αA-interferon expressed in E. coli have antiviral, antiproliferative, antineoplastic, and immunomodulating properties. Recombinant interferon αA (IFN-αA) is used as the first line of treatment for hepatitis C. It also displays a great therapeutic potential with chronic viral hepatitis, polycythemia, leukemia, and various other malignancies. It enhances natural killer cell activity, suppresses bone marrow function, and induces apoptotic cell death in non-melanoma skin cancer.

Unit Definition
One unit of interferon per ml of medium will cause a 50% inhibition of VSV-induced cytopathic effect on bovine MDBK cells. Units are determined with respect to the NIH international reference standard for human αA-IFN. Dilute in aqueous buffer containing at least 0.1% BSA or HSA.
Physical form
In PBS containing 0.1% BSA.
Preparation Note
Dilute only in buffers containing protein such as 0.1% BSA.
Reconstitution
Following initial thaw, aliquot and freeze (-70°C).
