Specificity

Clone 1G17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects BTG2. It targets an epitope within 21 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the internal region of human BTG2.

Application

Quality Control Testing

Evaluated by Western Blotting in Human Cerebellum tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected BTG2 in Human Cerebellum tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected BTG2 in Human brain tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected BTG2 in human cerebellum tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound BTG2 with a KD of 5.8 x 10-7 in an affinity binding assay.

Target description

Protein BTG2 (UniProt: P78543; also known as BTG family member 2, NGF-inducible anti-proliferative protein PC3) is encoded by the BTG2 (also known as PC3) gene (Gene ID: 7832) in human. BTG2 serves as an anti-proliferative protein whose expression is induced through a p53-dependent mechanism and its levels are significantly elevated in response to DNA damage. It plays a role in the negative control of cell proliferation and transfections of a BTG2 mammalian expression vector into NIH3T3 cells is reported to cause a decrease in their growth rate and cloning ability. BTG2 acts as a tumor suppressor and its levels are down-regulated in many human cancers. The low expression of BTG2 is found in many human cancer tissues and its loss is related to cell migration and invasion. BTG2 is reported to inhibit the proliferation and invasion of gastric cancer cells. Its overexpression can suppress the cell growth and promote apoptosis in pancreatic cancer cells. Its overexpression also reduces the levels of cyclin D1, MMP-1, and MMP-2, and suppresses the proliferation of lung cancer cells. BTG2 is one of the PRMT1 binding partner and its binding increases PRMT1 activity, which is essential for neurite outgrowth. BTG2 is phosphorylated at serine 147 by ERK1 and ERK2 and at serine 149 by MAPK14, leading to PIN1-binding and mitochondrial depolarization. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
