General description
CD107a (lysosome-associated membrane protein-1, LAMP-1), together with LAMP-2, is a major constituent of lysosomal membrane, 1-2% of total CD107a is found also on the plasma membrane. The LAMP proteins are involved in lysosome biogenesis and are required for fusion of lysosomes with phagosomes. Increased CD107a immunoreactivity is observed in neurones, and in glial cells surrounding senile plaques in Alzheimer′s disease cases and is localized mainly in medullary epithelial cells, single macrophages and lymphocytes in acute thymic involution. CD107a is a good marker of mast cell activation.
The gene cluster of differentiation 107a (CD107a), also known as lysosomal-associated membrane protein 1 (LAMP1), encodes a type I transmembrane protein. The protein has a short cytoplasmic tail containing a lysosome-targeting signal of GYQTI (382)-COOH. The gene is mapped to human chromosome 13q34.
Specificity
The antibody H4A3 recognizes CD107a, a 100-120 kDa glycoprotein expressed mainly on lysosomal, but also on the plasma membrane.

Application
The reagent is designed for Flow Cytometry analysis of human blood cells using 10 μL reagent / 100 μL of whole blood or 1e6 cells in a suspension. The content of a vial (1 mL) is sufficient for 100 tests.
Biochem/physiol Actions
The gene cluster of differentiation 107a (CD107a) encodes a membrane glycoprotein that functions as an intracellular receptor. It is found to be expressed in the cytoplasm of several types of tumor cells and may be involved in tumor invasion. CD107a is crucial for perforin trafficking to the lytic granules and motility of these lytic granules. Its knockdown leads to inhibition of cytotoxicity of human natural killer cells.
