General description

The pacFA lipid attached to 2-oleoyl-sn-glycero-3-phosphocholine, contains a photoactivable diazirine ring with a clickable alkyne group.

Biochem/physiol Actions

1-(9-(3-pent-4-ynyl-3-H-diazirin-3-yl)-nonanoyl)-2-oleoyl-sn-glycero-3-phosphocholine (pacFA-18:1 PC) may be used for global analysis of protein–lipid interactions in living cells. pacFA acts as a precursor for the biosynthesis of bifunctional lipids. Proteins in contact with the bifunctional lipids are then cross-linked by ultraviolet (UV) irradiation of the diazirine ring. Finally, click chemistry is used to label the alkyne with a reporter molecule.

Cell membranes contain hundreds to thousands of individual lipid species that are of structural importance but also specifically interact with proteins. Due to their highly controlled synthesis and role in signaling events sphingolipids are an intensely studied class of lipids.

Understanding biological processes at the mechanistic level requires a systematic charting of the physical and functional links between all cellular components. While protein-protein and protein-nucleic acid networks have been subject to many global surveys, other critical cellular components such as
