Specificity

Clone 2B7-L1 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects TIM-3/HAVCR2. It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human TIM-3/HAVCR2.

Application

Quality Control Testing

Evaluated by Western Blotting in Daudi cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TIM-3/HAVCR2 in Daudi cell lysate.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TIM-3/HAVCR2 in HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound TIM-3/HAVCR2 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Flow Cytometry Analysis: 1 µg from a representative lot detected TIM-3/HAVCR2 in one million human peripheral blood mononuclear cells (PBMC)..

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TIM-3/HAVCR2 in human tonsil tissue sections and human spleen tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Hepatitis A virus cellular receptor 2 (UniProt: Q8TDQ0; also known as HAVcr-2, T-cell immunoglobulin and mucin domain-containing protein 3 (TIMD-3), T-cell immunoglobulin mucin receptor 3 (TIM-3), T-cell membrane protein 3, CD366 ) is encoded by the HAVCR2 (also known as TIM3, TIMD3) gene (Gene ID: 84868) in human. As a member of the T cell immunoglobulin and mucin domain (TIM) family, TIM-3 is an important protein in the regulation of macrophage activation. TIM-3 is also critical for immune tolerance as it inhibits T-helper type 1 (Th1) mediated auto and alloimmune responses. It is a membrane bound protein and primarily localized on the plasma membrane of immune cells and also localizes to the immunological synapse between CD8+ T-cells and target cells. It is expressed on a variety of immune cells, including; T cells, dendritic cells, natural killer (NK) cells, macrophages, monocytes, and epithelial tissues. Expression is increased on CD4+ and CD8+ T-cells in chronic hepatitis C virus (HCV) infection. TIM-3 plays an important role in T-cell homing in the thymus and lymph and is the receptor for the Galectin-9 an important protein modulating cell-cell and cell matrix interactions. High TIM-3 expression is also associated with many tumors including renal cancer and acute myelogenous leukemia (AML), and its expression is also a target for anti-cancer therapy as anti-TIM-3 antibodies have been shown to eradicate leukemic stem cells (LSCs) in animal models of AML. Targeting Tim-3 kills two birds with one stone by balancing the immune system and eliminating leukemia stem cells (LSCs) in AML. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
