Specificity

Clone 2H2 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CD86. It targets an epitope within 16 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the extracellular domain of human CD86.

Application

Quality Control Testing

Evaluated by Western Blotting with MCF-7.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CD86 in MCF-7 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CD86 in human spleen tissue lysate and a 1:50,000 dilution detected T7/HIS-tagged, human recombinant CD86.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CD86 in human tonsil tissue sections.

Flow Cytometry Analysis: 1 µg from a representative lot detected CD86 in one million human peripheral blood mononuclear cells (PBMC).

Affinity Binding Assay: A representative lot of this antibody bound CD86 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD86 in HDLM-2 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

T-lymphocyte activation antigen CD86 (UniProt: P42081; also known as Activation B7-2 antigen, B70, BU63, CTLA-4 counter-receptor B7.2, FUN-1) is encoded by the CD86 (also known as CD28LG2) gene (Gene ID: 942) in human. CD86 is a type I transmembrane protein belonging to the immunoglobulin superfamily, containing an extracellular Ig-like V-type (aa 33-131) and Ig-like C2-type (aa 15-225) domains essential for T cell interaction, a transmembrane domain (aa 248-268) and a cytoplasmic domain. The extracellular domain of CD86 is responsible for binding to CD28 or CTLA-4 on T cells. CD86 is expressed on the surface of antigen-presenting cells, including dendritic cells, macrophages, and B cells. Its expression is upregulated upon activation of these cells, which typically occurs during an immune response to pathogens. Its dysregulation has been implicated in autoimmune diseases such as rheumatoid arthritis and multiple sclerosis, allergic reactions, and transplant rejection, making it a potential target for immunotherapy. In some cancers, the manipulation of CD86 and its pathways is explored for therapeutic cancer vaccines.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
