Specificity

Clone 5I3 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Integrin a6. It targets an epitope within 17 amino acids from the C-terminal half, extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal, extracellular domain of human Integrin a6.

Application

Quality Control Testing

Evaluated by Western Blotting in Cos-7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Integrin a6/CD49f in Cos-7 cell lysate.

Tested applications

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Integrin a6/CD49f in HeLa, A431, HUVEC, and NIH3T3 cells.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected Integrin a6/CD49f in one million human peripheral blood mononuclear cells (PBMC).

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Integrin a6/CD49f in human stomach tissue sections.

Anti-Integrin a6/CD49f, clone 5I3 ZooMAb®, Cat. No. ZRB1168, is a recombinant Rabbit monoclonal antibody that targets Integrin a6 and is tested for use in Flow Cytometry, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Integrin a6 (UniProt: P23229; also known as CD49 antigen-like family member F, VLA-6, CD49f) is encoded by the ITGA6 gene (Gene ID: 3655) in human. Integrins are heterodimeric integral membrane proteins composed of an a subunit and a b subunit that function in cell surface adhesion and signaling. They contain a large extracellular domain responsible for ligand binding, a single transmembrane domain, and a cytoplasmic domain. The exact combination of various a- and b-subunits dictates the binding specificity of integrins to different ECM components. Although both subunits are required for adhesion, the binding specificity primarily depends on the extracellular portion of the a-subunit. The structural and functional diversity of the integrin family is based upon the pairing abilities of the individual a and b subunits. Integrins are not constitutively active and their activation from a low ligand-binding affinity state to high ligand-binding affinity state requires conformational change that can originate from either from their cytoplasmic or extracellular interactions. The recognition site for most integrins that bind the ECM consists of an RGD (arginine-glycine-aspartic acid) sequence. Integrin a6 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 24-1050), a transmembrane domain (aa 1051-1076), and a cytoplasmic domain (aa 1077-1130). It can heterodimerize with b1 to form a receptor for laminin on platelets. a6/b1 is also present in oocytes and is involved in sperm-egg fusion. Integrin a6/b4 heterodimer is a receptor for laminin in epithelial cells and plays a role in the hemidesmosome. It also binds to IGF-1 and IGF-2 and these binding are shown to be essential for IGF signaling. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
