General description

STIM1 (stromal interaction molecule 1) is a transmembrane protein, which spans the membrane once. It predominantly resides in endoplasmic reticulum (ER), though some of it is found in the plasma membrane. It has a canonical EF-hand (cEF) domain, two EF-hand domains, a hidden non Ca2+ binding EF-hand domain (hEF), and a sterile α motif (SAM). This is present in the N-terminal of the protein, which is located towards the lumen of ER. This gene is localized to human chromosome 11p15.5, and encodes a protein of 90kDa molecular weight, which is ubiquitously expressed.

Application

Biochem/physiol Actions

STIM1 (stromal interaction molecule 1) is an integral part of SOCE (Store-operated Ca2+ entry), which is a Ca2+ entry gateway in non-excitable cells. Orai1, which is a Ca2+ channel present on plasma membrane, interacts with STIM1 present on ER, through SOAR/CAD domain present in the C-terminal of STIM1. Whenever Ca2+ levels fall low in ER, STIM1 changes to an extended conformation of C-terminal to expose SOAR/CAD domain. Also Ca2+ depletion leads to oligomerization of STIM1, through unfolding of the EF-SAM domain. STIM1 present on plasma membrane also regulates Ca2+ entry through opening of arachidonic acid calcium (ARC) channels. It acts as a tumor suppressor gene, and its expression is related to various malignancies. Studies show that up-regulation of this gene leads to tumor arrest in G401 rhabdoid tumor, rhabdomyosarcoma and rodent myoblast cell lines.
