Cell Line Description

H5 is a subclone of H4-II-E-C3 (Sigma Catalogue number. 85061112) which originated from the Reuber H-35 hepatoma induced by chemical carcinogenesis in an A x C rat. The dedifferentiated H5 cells express high levels of glucose-6-phosphate dehydrogenase and exhibit a population doubling time of 12 hours. Differentiated variants are H4IIEC3 (85061112), Fao and others.

Application

Toxicity studies; negative control for differentiated hepatoma

Culture Medium

Coon′s modified Ham′s F12 (or Kaign′s modified Ham′s F12 + 45mg/L ascorbic acid + 18mg/L Inositol) + 2mM Glutamine + 5% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:50 to 1:100 i.e. seeding at 1-2x1,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
