General description

mTOR (mammalian target of rapamycin) is a Ser/Thr kinase belonging to the phosphatidylinositol 3-kinase-related kinase protein family. It mediates cell growth, cell proliferation, cell motility, cell survival, protein synthesis and transcription in response to growth factors and energy needs. mTOR is part of the Akt/mTOR pathway which is an important pathway that controls cellular metabolism. Dysfunction of this pathway has been implicated in various human diseases such as diabetes, obesity and cancer. mTOR is encoded by the MTOR gene. The mTOR catalytic subunit exists in two structurally distinct complexes: mTORC1 and mTORC2. Each of the two complexes are composed of different interacting proteins and localized to different subcellular compartments which affect its activation and function. The core components of mTORC1 is composed of mTOR, regulatory-associated protein of mTOR (Raptor) and mammalian lethal with SEC13 protein 8 (MLST8). The core components of mTORC2 is composed of mTOR, rapamycin-insensitive companion of mTOR (RICTOR), MLST8, and mammalian stress-activated protein kinase interacting protein 1 (mSIN1).The MILLIPLEX® MAP 2-Plex Phospho/Total mTOR kit contains all the components necessary to measure total mTOR and phosphorylated mTOR (Ser2448). The MILLIPLEX® MAP 2-Plex Phospho/Total mTOR magnetic bead kit has been developed for the simultaneous detection of phosphorylated mTOR (Ser2448) and total mTOR in a single well using the Luminex system. This detection assay is a rapid, convenient alternative to Western Blotting and immunoprecipitation procedures for the analysis of cell lysate samples..

Specificity

Cross-reactivity between the antibodies and any of the other analytes in this panel is non-detectable or negligible.

Application

The MILLIPLEX® 2-Plex Phospho/Total mTOR kit detects total mTOR and phosphorylated mTOR (Ser2448).An overnight (4°C) incubation is recommended for best results.This assay requires 25 μL diluted, filtered cell lysate per well.This kit must be run using the provided Assay Buffer.The suggested working range of protein concentration for the assay is 1 to 25 μg of total protein/well (25 μL/well at 40 to 1,000 μg/mL)Analytes available:mTOR (Total)mTOR (Ser2448)
