Biochem/physiol Actions
MF-094 is a potent inhibitor of ubiquitin specific protease 30 (USP30), a de-ubiquitinating enzymes (DUB) that opposes the action of parkin-mediated ubiquitination in clearing damaged mitochondria. MF-094 is selective for USP30 with an IC50 value of 120 nM, compared to >10 μM for USPs 1, 8, and 9.

Mitophagy is one of the processes that cells use to maintain overall health. An E3 ligase, parkin, ubiquitinates mitochondrial proteins prior to their degradation by autophagasomes. USP30 is an enzyme that de-ubiquitinates mitochondrial proteins; therefore, inhibiting this enzyme could foster

The death of cardiomyocytes either through apoptosis or necroptosis is the pathological feature of cardiac ischemia-reperfusion (I/R) injury. Phosphoglycerate mutase 5 (PGAM5), a mitochondrially-localized serine/threonine-protein phosphatase, functions as a novel inducer of necroptosis. However, intense debate exists regarding the effect
