Gene Information

human ... ALOX5(240)
rat ... Alox5(25290)

General description

The gene ALOX5 (arachidonate 5-lipoxygenase), also referred to as 5-LOX (5-lipoxygenase), encodes a protein belonging to the lipoxygenase family. The gene is mapped to human chromosome 10q11.21. The protein contains a distinct ‘C2-like′ domain at the amino terminal that is involved in Ca2+-dependent membrane binding and a large catalytic domain containing the non-heme catalytic iron. It is highly unstable with short-lived activity.

Immunogen

phosphopeptide corresponding to amino acid residues surrounding the pSer523 of human 5-lipoxgenase (5-LO).

Biochem/physiol Actions

The gene ALOX5 (arachidonate 5-lipoxygenase) encodes a lipoxygenase that catalyzes the conversion of arachidonic acid (AA) and other fatty acids to bioactive compounds, such as 5(S)-hydroxyeicosatetraenoic acid (5-HETE), leukotriene B4 (LTB4), and other leukotrienes. These leukotrienes and lipoxins function as mediators of inflammatory response. ALOX5 has been implicated in tumorigenesis and may serve as a potential therapeutic target and biomarker.

Physical form

Solution in 10 mM HEPES, pH 7.5, 150 mM NaCl, 100 μg/mL BSA, and 50% glycerol.

Leukotrienes are inflammatory mediators generated from arachidonic acid (polyunsaturated n-6 fatty acid) by the enzyme 5-lipoxygenase. Since atherosclerosis involves arterial inflammation, we hypothesized that a polymorphism in the 5-lipoxygenase gene promoter could relate to atherosclerosis in humans and that this
ALOX5 polymorphism associates with increased leukotriene production and reduced lung function and asthma control in children with poorly controlled asthma.

Eicosanoids are inflammatory mediators that play a key but incompletely understood role in linking the innate and adaptive immune systems. Here, we show that cytotoxic effector T cells (CTLs) are capable of both producing and responding to cysteinyl leukotrienes (CystLTs)
