General description

SLC9A3R1 (solute carrier 9A3 receptor 1), commonly known as Na+/H+ exchanger regulatory factor 1 (NHERF1), is an ezrin-radixin-moesin (ERM)-binding phosphoprotein 50. This gene is localized to human chromosome 17q25.1, and codes for a scaffold protein. This protein consists of two tandem PDZ domains and a C-terminal ezrin-binding region.

Application

Biochem/physiol Actions

SLC9A3R1 (solute carrier 9A3 receptor 1) protein plays an essential role in the binding of integral membrane proteins to cytoskeleton. Thus, it is thought to be a key player in cell signaling involved in cytoskeleton dynamics. Its expression is linked with multiple types of cancer and their aggressiveness. In breast cancer, its loss of nuclear expression is linked with decreased survival. Studies in colon adenocarcinoma HT-29 epithelial cells show that this protein plays an essential role in cellular proliferation through Gq-mediated signalling pathways. It also controls EGF (epidermal growth factor) and neurotensin signaling in HT-29 cell line. It plays an essential role in the biogenesis of microvilli, where it is critical for the retention of specific proteins in the microvilli. Activation by Rac1 protein results in conformation changes in SLC9A3R1 protein, which leads to the anchoring of cystic fibrosis transmembrane conductance regulator (CFTR) in the cell membrane.
