Cell Line Description

From parental strain L which was derived from normal subcutaneous areolar and adipose tissue of 100 day old male C3H/An. Used in development of replicate culture techniques, chemically defined media, fluid suspension culture methods and virus assays. Susceptible to pseudorabies virus and vesicular stomatitis (Indiana strain) virus.

Application

Replication, biochemical, neoplastic transformation, nutritional, metabolic enzyme, morphological, chromosome, immunologic, radiation, toxicologic and virus studies

Culture Medium

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:10 i.e. seeding at 1-3x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C. Freeze in 10% glycerol/90% FBS. After resuscitation cells may take up to 4 days to establish growth.
