General description

Assurance® GDS is an advanced genetic detection system for food pathogens. The MPX ID for Top STEC is a secondary automated nucleic acid amplification system for the identification of “Top Six” non-O157 Shiga Toxigenic E. coli (STEC). The Top 6 non-O157 STEC are defined as E. coli belonging to serogroups O103, O111, O121, O145, O26, or O45 that possess both the eae gene and at least one of the Shiga Toxin genes (stx1 or stx2). GDS MPX ID utilizes a proprietary PickPen® IMS-based sample preparation procedure to quickly and effectively capture organisms belonging to the 6 specific Top STEC O-serogroups prior to genetic analysis for the associated pathogenicity genes. Each of the 3 unique eae gene sequences targeted by MPX ID are highly correlated plus one or more of the 6 O-serogroups comprising Top 6 non-O157 STEC. Because there are 3 different eae gene targets and 6 different O-serogroup targets, the PCR reactions have been divided between 2 Amplification Tubes. Each tube contains an internal control and all necessary PCR reagents. Results will automatically be combined between the 2 tubes via GDS Rotor-Gene® software.

Application

Assurance® GDS MPX ID for Top STEC is designed to follow GDS MPX Top 7 STEC (PN 71015BC) for the identification of “Top Six” non-O157 Shiga Toxigenic E. coli (STEC) in a variety of foods, ingredients, and environmental samples. Additionally, GDS MPX ID can be used to confirm isolated colonies from samples presumptive positive for Top 6 STEC.
