General description

DNA replication protein Cdc45 (UniProt: Q4U3P7; also known as Cdc45) is encoded by the CDC45 gene (Gene ID: 100689362) in hamster. Cdc45 is a replication initiation protein that plays an essential role in the establishment and progression of the replication fork. It is required for formation of pre-initiation complexes at the G1/S transition and for activation of replication origins. Cdc45 associates with the heterohexameric Mcm2-7 helicase and the GINS complex (go-ichi-nii-san, Japanese for 5-1-2-3, for the subunits Sld5, Psf1, Psf2 and Pfs3) to form the CMG (Cdc45-Mcm2-7-GINS) complex. CMG represents the eukaryotic replicative DNA helicase that unwinds origins of replication and drives the replication fork. Cdc45 is also reported to interact with several other replication proteins, including DNA helicase B (DHB), Mcm10, Orc2, TopPB1, and DNA polymerases alpha, delta and epsilon. Expression of Cdc45 is associated with the proliferative status of the cell or tissue. Terminally differentiated and senescent cells do not display Cdc45 activity whereas highly proliferative cells express high levels of Cdc45.

This rabbit polyclonal antibody detects Cdc45 in Human and Murine cells.

Application

Anti-Cdc45, Cat. No. ABE2902, is a highly specific rabbit polyclonal antibody that targets DNA replication protein Cdc45 and has been tested for use in Western Blotting.
Western Blotting Analysis: A 1:500 dilution from a representative lot detected Cdc45 in 10 µg of HeLa cell lysate.

Western Blotting Analysis: A representative lot detected Cdc45 in asynchronous, go synchronized and G1 released CHO cells, immunoprecipitated CDC45 from CHO cells, HeLa and NIH3T3 (Wong, P.G., et. al. (2011). PLoS One. 6(3):e17533).

Quality

Evaluated by Western Blotting in CHO cell lysate.

Western Blotting Analysis: A 1:500 dilution of this antibody detected Cdc45 in 10 µg of CHO cell lysate.

~65 kDa observed; 65.36 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
