Application

BAM8-22 trifluoroacetate salt has been used in the study to identify the functions of Mas-related gene (Mrg) C in the pathogenesis and treatment of bone cancer pain (BCP).

Biochem/physiol Actions

BAM8-22 is a selective agonist for the sensory neuron specific G-protein coupled receptors (SNSRs). BAM8-22 inhibits development of morphine tolerance in rats. BAM8-22 is inactive at opioid receptors. Also, BAM8–22 is a potent activator of Mas-related G-protein-coupled receptors (Mrgprs), MrgprC11 and hMrgprX1. BAM8-22 produces itch and nociceptive sensations in humans in a histamine-independent manner.
Bovine adrenal medulla 8-22 (BAM8-22), a 15-amino acid peptide, acts a cleavage product of proenkephalin A and the precursor of Leu- and Met-enkephalin in the adrenal medulla. It may also be used to treat pain. It blocks inflammatory pain, chemical pain and downregulates the expression of spinal c-fos gene in an opioid-independent manner. In mice, BAM8-22 diminishes bone cancer pain. It induces the expression of Mas-related G-protein-coupled receptor C (MrgC) in the spinal dorsal horn.
