General description

Metabolic syndrome is a cluster of conditions that occur together; it includes increased blood pressure, elevated insulin levels, excess body fat around the waist and abnormal cholesterol levels.Key features of metabolic syndrome include insulin resistance, glucose intolerance, hypertension, dyslipidemia and central obsesity- all of which are risk factors for atherosclerosis, coronary heart disease, type 2 diabetes, kidney disease, and even premature death.Adults with metabolic syndrome show a low-grade inflammation, whose link with obesity may be dysregulated adipocyte production of pro- and anti-inflammatory factors. Consequently, research done in this area covers multi-faceted fields of cytokines, acute phase proteins, diabetes, and obesity related hormones, as well as other cardiovascular disease biomarkers.The MILLIPLEX® Mouse Metabolic Hormone Expansion Panel is a 15-plex kit to be used for the simultaneous quantification of any or all of the following analytes in tissue/lysate and culture supernatant samples and mouse serum or plasma: Amylin Active, C-Peptide 2, Ghrelin, GIP, GLP-1 Active or GLP-1 Total, Glucagon, IL-6, Insulin, Leptin, MCP-1, PP, PYY, Resistin, Secretin, and TNFα.Note: GLP-1 Active an GLP-1 Total cannot be run together in the same assay.

Specificity

Cross-reactivity between the antibodies and any of the other analytes in this panel is non-detectable or negligible.

Application

The Mouse Metabolic Hormone Expanded Panel, using the Luminex® xMAP technology, enables the simultaneous analysis of 15 protein biomarkers in mouse serum, plasma and tissue/lysate and culture supernatant samples.There are 16 biomarkers to choose from in this kit. However, this is a 15-plex kit since the following analytes cannot be run simultaneously: GLP-1 (Active) & GLP-1 (Total).The following analytes require the addition of protease inhibitors to the serum or plasma samples (see sample collection and storage section in the protocol for details): GLP-1 (DPPIV inhibitor); Amylin (protease inhibitor cocktail); Glucagon (Aprotinin); Ghrelin (Active) (serine protease inhibitor). The protocol recommends serum or plasma samples be run neat. Tissue culture samples may be run neat or may require a dilution with an appropriate control medium. For optimal sensitivity, use an overnight (16 - 18 hour) incubation.
