General description
The selectivity of the medium is contributed using antibiotics- Cefsulodin, Irgasan(Triclosan), Novobiocin, bile salts and crystal violet dye which synergistically inhibit the accompanying flora. The pyruvate in the medium promotes the growth of Yersinia spp. agar is the solidifying agent.
Mannitol from the medium is degraded by the growing Yersinia to form an acid which will trigger a pH change indicated by neutral red. The positive colonies appear red in color. The fermentation reaction by the growing Yersinia is intense and localized which produces the characteristic “bull′s eye” appearance. The microbes which do not metabolize mannitol from the media will form colorless, translucent colonies.
Application
Analysis Note
- The antibiotics- Cefsulodin, Irgasan(Triclosan), Novobiocin are added separately after autoclaving.
- The dehydrated medium is granulate with pink color.
- The prepared medium is clear to slightly opalescent and red.
