Tryptic Soy Broth

Tryptic Soy Broth, commonly referred to as Tryptone soya broth or soybean casein digest broth, is a liquid medium utilized in microbiology. Its primary purposes include detecting sterility, cultivating fastidious microorganisms, and enriching them. The broth comprises enzymatic digests of soybean meal and casein, peptones that provide amino acids and nitrogenous compounds. Additionally, it contains sodium chloride to maintain osmotic equilibrium, and dextrose, which provides carbohydrates and energy. This medium also includes vital vitamins and minerals, which are important for the growth of organisms. Dipotassium hydrogen phosphate is included as a buffering agent to maintain the pH balance. Tryptic Soy Broth is used to culture a wide range of aerobic gram-positive and gram-negative bacteria, fungi, routine blood cultures, and microbial biofilms.
Tryptic Soy Broth in Pathogen Testing
CASO Broth, also known as casein-peptone soymeal-peptone broth, is a variation of tryptic soy broth. It is utilized to examine the sterility of products or preparations that are not guaranteed to be sterile. They are used to identify specified microorganisms and total viable aerobic counts through the most probable number (MPN) method. These broths function as pre-enrichment media for detecting foodborne pathogens like Salmonella and Shiga toxin-producing E. coli.
Tryptic Soy Broth in Sterility Testing
Tryptic soy broth with neutralizers TLHC (containing tween® 80, lecithin, histidine, and cystine), 2x concentrated tryptic soy broth, or tryptic soy broth with tween® 80 are useful for isolating and cultivating delicate bacteria and yeast.

GranuCult®, a modified tryptic soy broth (mTSB) is used to enrich E. coli and Salmonella species in red meats. The use of mTSB media effectively inhibits the growth of other gram-negative Enterobacteriaceae species by 90%. GranuCult® with Novobiocin is used for enriching enterohemorrhagic E. coli (EHEC) and detecting E. coli serotype O157:H7 in food samples. The mTSB broth selective supplement contains lyophilized Novobiocin sodium salt, which can effectively suppress the growth of concurrent Gram-positive bacteria during the cultivation of E. coli (O157:H7).
