Specificity

Clone 1I17 is a ZooMAb® recombinant rabbit monoclonal antibody that detects LGR5/GPR49. It targets an epitope within 25 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the extracellular domain of human LGR5/GPR49.

Application

Quality Control Testing

Evaluated by Western Blotting in HL-60 cell lysate.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected LGR5/GPR49 in HL-60 cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound LGR5/GPR49 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected LGR5/GPR49 in human colon tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected LGR5/GPR4 in SH-SY5Y cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Leucine-rich repeat-containing G-protein coupled receptor 5 (UniProt: O75473; also known as G-protein coupled receptor 49, G-protein coupled receptor 67, G-protein coupled receptor HG38) is encoded by the LGR5 (also known as GPR49, GPR67) gene (Gene ID: 8549) in human. It is a G-protein coupled receptor (GPCR) characterized by an extracellular domain containing multiple leucine-rich repeat (LRR) sequences, critical for protein-protein interactions. The structure also includes a seven-transmembrane domain typical of GPCRs. LGR5/GPR49 is a receptor for R-spondins that potentiates the Wnt signaling pathway, a key regulator of cell proliferation, differentiation, and migration, stem cell renewal and growth. This receptor is crucial in maintaining the stem cell population in various tissues, particularly in the intestinal crypts where it supports rapid turnover and regeneration. Overexpression of LGR5/GPR49 has been observed in various cancer types, where it is associated with tumor growth, metastasis, and poor prognosis. It is a potential marker for identifying cancer stem cells within tumors, which are implicated in resistance to conventional therapies and in the recurrence of cancer.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
