Specificity

Clone 1E16 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects MHC class II antigen DPB1 (HLA-DPB1). It targets an epitope within 18 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the extracellular domain of human MHC class II antigen DPB1 (HLA-DPB1).

Application

Quality Control Testing

Evaluated by Western Blotting in Human tonsil tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HLA-DPB1 in Human tonsil tissue lysate.

Tested Applications

Flow Cytometry Analysis: 1 µg from a representative lot detected HLA-DPB1 in one million Human peripheral blood mononuclear cells (PBMC).

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HLA-DPB1 in HEK 293 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HLA-DPB1 in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound HLA-DPB1 peptide with a KD of 1.0 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

HLA class II histocompatibility antigen, DP beta 1 chain (UniProt: P04440; also known as HLA class II histocompatibility antigen, DP(W4) beta chain, MHC class II antigen DPB1) is encoded by the HLA-DPB1 (also known as HLA-DP1B) gene (Gene ID: 3115) in human. HLA-DPB1 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-29), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 30-225), a transmembrane domain (aa 226-246), and a cytoplasmic domain (aa 227-258). It plays a central role in the immune system by presenting peptides derived from extracellular proteins. It is a MHC class II molecule that forms a heterodimer of α and β subunits. Its expression is observed in antigen presenting cells. It is reported to bind peptides derived from antigens that ACCESS the endocytic route of antigen presenting cells (APC) and present them on the cell surface for recognition by the CD4 T-cells. Its peptide binding cleft accommodates peptides of 10-30 residues. Endocytosed exogenous antigens are readily available for presentation and compete with endogenous proteins on their way to degradation in lysosomes. Genetic variants in HLA-DPB1 are associated with increased risk of severe aplastic anemia. A mutation in HLA-DPB1 gene (HLA-DPB1Glu69) is observed in chronic beryllium disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
