Specificity

Clone 5K9 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Interleukin-33.

Immunogen

GST/His-tagged recombinant fragment corresponding to 146 amino acids from the C-terminal region of human IL-33.

Application

Anti-IL-33, clone 5K9 ZooMAb, Cat. No. ZRB1350, is a recombinant Rabbit monoclonal antibody that specifically targets IL-33 and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), an Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IL-33 in rat spleen, human spleen, EL4, and RAW264.7 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected IL-33 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IL-33 in RAW 264.7 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interleukin-33 (UniProt: O95760; also known as IL-33, Interleukin-1 family member 11, IL-1F11, Nuclear factor from high endothelial venules, NF-HEV) is encoded by the IL33 (also known as C9orf26, IL1F11, NFHEV) gene (Gene ID: 90865) in human. IL-33 is a tissue-derived nuclear cytokine of the IL-1 family that is highly expressed in endothelial cells, epithelial cells and fibroblast-like cells. It is normally released from damaged and necrotic barrier cells and serves as an alarmin to promote activation of the innate immune system and removal of pathogens. It is considered as a ubiquitous immune modulator. IL-33 is expressed at high levels in endothelial venules in tonsils, Peyer patches, and mesenteric lymph nodes. It exerts its cytokine activity by forming a 1:1:1 heterotrimeric complex with its primary high-affinity receptor IL1RL1 and the co-receptor IL1RAP. This complex activates NF-kappa B and MAP kinase signaling in target cells. IL-33 is also reported to be involved in the maturation of Th2 cells and inducing the secretion of T-helper type 2-associated cytokines. Although full-length IL-33 can signal via IL1R1 receptor, proteolytic processing by cathepsin G and neutrophil elastase produces C-terminal peptides that are shown to be more active than the unprocessed full-length protein. IL-33 can also undergo proteolytic cleavage by caspase-3 and -7 that results in its activation. IL-33 has a homeodomain-like HTH domain (aa 1-65) that mediates its nuclear localization and heterochromatin association. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
