General description

VAPA (vesicle associated membrane protein associated protein A) gene is localized to human chromosome 18p11. The encoded protein is also called VAP33, which shows a wide range of tissue expression. It is an integral membrane protein, which is composed of coiled-coil domains. This protein was originally isolated from Aplysia as an interacting partner of VAMP/synaptobrevin. This protein co-localizes with occluding at the tight junctions in cells. This protein has a molecular weight of 33kDa.

Application

Biochem/physiol Actions

VAPA (vesicle associated membrane protein associated protein A) plays an essential role in vesicular neurotransmission, where it interacts with presynaptic proteins. Studies suggest that SNPs (single nucleotide polymorphisms) in the 3′UTR (untranslated region) of this gene are linked with bipolar disorder. It is essential for the replication of HCV (hepatitis C virus), where it interacts with HCV nonstructural protein 5A (NS5A) in a G protein pathway suppressor 2 (GPS2)-mediated manner. As this protein binds with VAMP/synaptobrevin, it is thought to be involved in vesicle docking and fusion.
