Spécificité

Clone 1G7 is a ZooMAb® rabbit recombinant monoclonal antibody that detects YB1. It targets an epitope within 23 amino acids from the N-terminal region.

KLH-conjugated linear peptide corresponding to 23 amino acids from the N-terminal of human YB1.

Application

Quality Control Testing

Evaluated by Western Blotting in T-47D cell lysate.

Western Blotting Analysis: A 1:25,000 dilution of this antibody detected YB1 in T-47D cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected YB1 in C6 and human testis lysates.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected YB1 in HEK-293 cells.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected YB1in HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected YB1 in Human testis tissue sections.

Flow Cytometry Analysis: 0.1 µg of this antibody detected YB1 in one million fixed and permeabilized HeLa cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Y-box-binding protein 1 (UniProt: P67809; also known as YB-1, CCAAT-binding transcription factor I subunit A (CBF-A), DNA-binding protein B, Enhancer factor I subunit A, EFI-A, Nuclease-sensitive element-binding protein 1, Y-box transcription factor) is encoded by the YBX1 (also known as NSEP1, YB1) gene (Gene ID: 4904) in human. Y-box-binding protein 1 (YB-1) belongs to the cold shock domain (CSD) protein family, which is characterized by a highly conserved nucleic acid-binding domain. YB-1′s structure includes the CSD at the N-terminus and a diverse C-terminal domain that contains both alternating basic and acidic regions. This structure facilitates its role in binding to both DNA and RNA, enabling a wide range of functions. YB-1 is a multifunctional protein that plays significant roles in various cellular processes including regulation of transcription, translation of mRNAs, DNA repair, and stress response. It acts as a transcription factor that can both activate and repress genes. In translation, YB-1 binds to mRNA and can affect its stability and translation efficiency, often in response to cellular stress. YB-1 is ubiquitously expressed across various tissues but shows higher expression levels in lung, colon, and breast tissues. Its overexpression has been observed in many cancers, including breast, ovarian, lung, and prostate cancers, and is often linked with aggressive tumor growth, increased metastasis, and resistance to treatment. Additionally, YB-1 has been implicated in other pathological conditions such as inflammation and fibrosis.

Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
