Spécificité

Clone 1C10 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects TSC1. It targets an epitope within 20 amino acids from the C-terminal half.

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in human skeletal muscle tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution of this antibody detected TSC1 in human skeletal muscle tissue sections.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected TSC1 in human cerebellum tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TSC1 in A431 cells.

Affinity Binding Assay: A representative lot of this antibody bound TSC1 with a KD of 4.5 x 10-7 in an affinity binding assay.

Description de la cible

Hamartin (UniProt: Q92574; also known as Tuberous sclerosis 1 protein, TSC1) is encoded by the TSC1 (also known as KIAA0243, TSC) gene (Gene ID: 7248) in human. TSC1 is a peripheral membrane protein that is highly expressed in skeletal muscle, followed by heart, brain, placenta, pancreas, lung, liver, and kidney. Its expression is also observed in embryonic kidney cells. In complex with TSC2, it inhibits the nutrient-mediated or growth factor-stimulated phosphorylation of S6K1 and eukaryotic translation initiation factor 4E-binding protein 1 (EIF4EBP1) by negatively regulating mTORC1 signaling. It acts as a co-chaperone for HSP90AA1 facilitating HSP90AA1 chaperoning of proteins, such as kinases, TSC2 and glucocorticoid receptor NR3C1. It increases ATP binding to HSP90AA1 and inhibits HSP90AA1 ATPase activity. It is reported to recruit TSC2 to HSP90AA1 and stabilize TSC2 by preventing the interaction between TSC2 and HECT and RLD domain containing E3 ubiquitin ligase (HERC1). It contains a C-terminal putative coiled-coil domain (721-997), which is shown to be essential for its interaction with TSC2. TSC1 and TSC2 form a protein complex (TSC1/2) that negatively regulates the mTOR pathway by inactivating RHEB. Both TSC1 and TSC2 are reported to be necessary for a functional TSC1/2 complex and loss of either gene can leads to the clinical manifestations of TSC and loss of either TSC1 or TSC2 leads to unchecked activation of mTOR. Hence, TSC1 can serve as a tumor suppressor. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
