Specificity
Clone 2D12 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects PAX7. It targets an epitope within 25 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of human Paired box protein Pax-7 (PAX7).
Application
Quality Control Testing
Evaluated by Immunohistochemistry (Paraffin) in Human testis tissue sections.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected PAX7 in Human testis tissue sections.
Tested Applications
Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected GST/His-tagged recombinant Human PAX7.
Affinity Binding Assay: A representative lot of this antibody bound PAX7 peptide with a KD of 1.4 x 10-7 in an affinity binding assay.
Flow Cytometry Analysis: 1 µg from a representative lot detected PAX7 in one million HeLa cells.

Target description
Paired box protein Pax-7 (UniProt: P23759; also known as HuP1, PAX7) is encoded by the PAX7 (also known as HUP1) gene (Gene ID: 5081) in human. PAX7 is a transcription factor that is involved in the regulation of muscle stem cells proliferation and plays a role in myogenesis and muscle regeneration. It can bind to DNA as a heterodimer with PAX3. Its homeobox DNA-binding domain is localized in amino acids 217-276. PAX7 has two acetylation sites (Lys 105 and Lys 193) that positively regulate its transcriptional activity and lack of acetylation reduces its DNA binding. Its acetylation levels are regulated by MYST1 and SIRT2. PAX7 expression is observed in quiescent as well as activated satellite cells, and its levels are downregulated when satellite cells commit to muscle differentiation. It is also critical for the normal function of satellite cells in adult skeletal muscle. PAX7 deficiency results in cell-cycle arrest and early differentiation. A chromosomal aberration involving PAX7 is found in cases of rhabdomyosarcoma, a highly malignant tumor of striated muscle derived from primitive mesenchymal cells. Three isoforms of PAX7 have been described that are produced by alternative splicing. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
