Specificity

Clone 4D5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Cyclophilin B. It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Cyclophilin B.

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Cyclophilin B in NIH3T3 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Cyclophilin B in lysates from COS-7, C6, A431, and U87 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Cyclophilin B in human thyroid tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound recombinant Human Cyclophilin B with a KD of 3.6 x 10-6 in an affinity binding assay.

Enzyme Immunoassay Analysis (ELISA): A representative lot of this antibody detected recombinant Human Cyclophilin B in ELISA application.

Target description

Peptidyl-prolyl cis-trans isomerase B (UniProt: P23284; also known as EC:5.2.1.8, PPIase B, CYP-S1, Cyclophilin B, Rotamase B, S-cyclophilin, SCYLP) is encoded by the CYPB (also known as PPIB) gene (Gene ID: 5479) in human. Cyclophilins are a group of peptidyl-prolyl cis/trans isomerases that play crucial role in regulatory mechanisms of cellular physiology and pathology in several inflammatory conditions. Cyclophilin B (CypB) is an endoplasmic reticulum (ER) resident protein that is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to produce mature active protein (aa 34-216). It catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides and therefore assists in protein folding. CypB is secreted by several cell types, and it possesses proinflammatory properties. It is reported to play a key role in ER redox homeostasis. Higher levels of CypB are observed in inflammatory processes, like sepsis and rheumatoid arthritis. It also promotes platelet adhesion to collagen and the neutrophil chemotaxis. It binds to the CD147 membrane receptor to modulate inflammatory pathways. CypB is reported to provide essential survival signals in glioblastoma multiforme cells. Suppression of CypB is shown to reduce the proliferation of glioblastoma cells. Depletion or pharmacological inhibition of CypB causes hyperactivation of the oncogenic RAS-mitogen-activated protein kinase pathway, induction of cellular senescence signals, and death resulting from loss of MYC, mutant p53, Chk1, and JAK/STAT3 signaling. Mutations in CYPB gene are known to cause Osteogenesis imperfecta, a disease characterized by low bone mass, bone fragility and increased susceptibility to fractures even after minimal trauma. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
