General description

Human Interferon alpha-2 (IFNa2) is a cytokine belonging to the IFN family exhibiting antiviral, antitumor and antiproliferative effects on immune response. IFNa2 is secreted by viral infected cells and acts to inhibit viral infection. It has been shown to play a role in autoimmune diseases such as lupus and myositis. The mechanisms of action underlying the pathogenic role that IFNa and IFNß play in autoimmunity is not well understood but elucidating their role will lead to a better understanding of these cytokines in chronic inflammation and immunity.

The SMC® Human IFN-α2 High Sensitivity Immunoassay Kit contains all reagents required to perform the assay. This assay takes advantage of a standard immunoassay workflow configured in a standard plate format.
Assay protocols are similar to existing sandwich ELISA methods with two key differences:
1) Elution buffer disrupts the sandwich, separating the labeled detection antibody for quantification.
2) The Erenna and SMCxPRO® immunoassay systems detect analytes using Single Molecule Counting (SMC®) technology.

Specificity

Cross Reactivity:
This assay was tested for cross reactivity with rhIFN-β, rhIFN-γ, rhIFN-λ1 (IL-29), rhIFN-λ2 (IL28A), rhIFN-λ3 (IL-28B), rhIFN-ω, IFN-α2a, IFN-α2b and IFN-α.

  • There is no cross-reactivity detected with rhIFN-ß, rhIFN-γ, rhIFN-λ1 (IL-29), rhIFN-λ2 (IL-28A),rhIFN-λ3 (IL-28B), and rhIFN-ω.
  • There is 100% cross-reactivity with IFN-α2a.
  • There is 49% cross-reactivity with IFN-α2b.
  • There is 3% cross-reactivity with IFN-α.

Accuracy:
  • Minimum of 5 serum samples spiked with average recovery of 102% with a range of 92-111%
  • Minimum of 5 plasma samples spiked with average recovery of 96% with a range of 89-107%
