General description

Phosphatidic acid (PA) is an anionic cellular glycerophospholipid constituting 1-4% of membrane lipids. It possesses a phosphomonoester polar head group and two long hydrophobic acyl chains. NBD PA is a fluorescently labelled lipid with the fluorophore, 7-nitrobenz-2-oxa-1,3-diazol-4-yl (NBD) at the sn-2 fatty acid of PA.

Application

18:1-12:0 NBD PA or 1-oleoyl-2-{12-[(7-nitro-2-1,3-benzoxadiazol-4-yl)amino]dodecanoyl}-sn-glycero-3-phosphate (ammonium salt) is suitable: for the in vitro enzyme activity assay of galactolipase, for NBD-PA and NBD phosphatidyl choline uptake assays, to measure lipid transfer specificity of Ups1 mutants

Biochem/physiol Actions

Phosphatidic acid (PA) is implicated in many membrane fusion and membrane fission events, such as fission of Golgi membranes. It is a bioactive lipid and a secondary messenger.

Phospholipases play crucial roles in plant membrane lipid homeostasis. Nonspecific phospholipase C (NPCs) establish a unique class of phospholipases found only in plants and certain bacteria. Here, we show that two previously uncharacterized NPC isoforms, NPC2 and NPC6, are required

Cholesterol plays a pivotal role in mitochondrial steroidogenesis, membrane structure, and respiration. Mitochondrial membranes are intrinsically low in cholesterol content and therefore must be replenished with cholesterol from other subcellular membranes. However, the molecular mechanisms underlying mitochondrial cholesterol transport remains

Phosphatidic acid (PA) is the simplest cellular glycerophospholipid characterized by unique biophysical properties: a small headgroup; negative charge; and a phosphomonoester group. Upon interaction with lysine or arginine, PA charge increases from -1 to -2 and this change stabilizes protein-lipid
