General description

Neurogenin-2 (UniProt P70447; also known as Helix-loop-helix protein mATH-4A, mATH4A, NGN-2, Protein atonal homolog 4) is encoded by the Neurog2 (also known as Ath4a, Atoh4, Ngn2) gene (Gene ID 11924) in murine species. Neuronal differentiation is largely under the control of basic Helix-Loop-Helix (bHLH) proneural transcription factors that play key roles during development of the embryonic nervous system. Neurogenin-2 (NGN-2) is a bHLH proneural transcription factor that orchestrates the progression from neural progenitor to differentiating neuron in several regions of the central nervous system, including the formation of sensory neurons from the epibranchial placode and neurogenesis in dorsal root ganglia. NGN-2 dimerizes with TCF3 (E12/E47), the heterodimer then binds to target DNA regions and recruit transcriptional coactivators, such as p300/CBP and Brg1, to facilitate the transcription of a cascade of additional bHLH transcription factors, such as NeuroD, as well as structural and functional genes associated with mature neuronal activity.

Specificity

The specific staining by clone 7G4 was blocked by NGN2-GST fusion, but not by NGN1-fusion protein (Lo, L., et al. (2002). Development. 129(7):1553-1567).

Application

Anti-Neurogenin-2, clone 7G4, Cat. No. MABN1814, is a highly specific mouse monoclonal antibody that targets Neurogenin-2 and has been tested in Immunocytochemistry, Immunofluorescence, and Immunohistochemistry (Paraffin).
Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected neurogenin-2 in mouse small intestine and colon tissue sections.

Immunocytochemistry Analysis: A representative lot detected NGN2-positive cells in dissociated neural tube (dNT) cultured under 10 ng/mL BMP2 for 48 hrs by fluorescent immunocytochemistry (Lo, L., et al. (2002). Development. 129(7):1553-1567).

Immunofluorescence Analysis: A representative lot detected NGN2 immunoreactivity among a small subset of cells at the dorsal margin of the neural tube and among a greater number of SOX10-positive cells in the neural crest migration pathway by fluorescent immunohistochemistry staining of 4% paraformaldehyde-fixed E10.5 rat spinal cord section (Lo, L., et al. (2002). Development. 129(7):1553-1567).

Quality

Evaluated by Immunohistochemistry in mouse cerebral cortex tissue section.

Immunohistochemistry Analysis: A 1:50 dilution of this antibody detected neurogenin-2 in mouse cerebral cortex tissue section.
