Target description

UL16-binding protein 2 (UniProt: Q9BZM5; also known as ALCAN-alpha, NKG2D ligand 2, N2DL-2, NKG2DL2, Retinoic acid early transcript 1H) is encoded by the ULBP2 (also known as N2DL2, RAET1H, UNQ463/PRO791) gene (Gene ID: 80328) in human. The UL16-binding proteins (ULBPs) belong to the family of MHC class I-related molecules that are targets of human CMV glycoprotein. ULBP2 is a GPI-anchor glycoprotein that is synthesized with a signal peptide (aa 1-25) and a propeptide (aa 217-246), which are subsequently cleaved off to produce the mature form. ULBP2 serves as a ligand of the activating immunoreceptor NKG2D and mediates natural kill (NK) cell cytotoxicity. It is detected in cell membrane and endoplasmic reticulum. A secreted for has also been described. Its expression is observed in cancer cells and in the fetus, but not in normal tissues. Tumor cells are reported to release the soluble form of ULBP2 by the action of metalloproteases, however, unlike membrane bound form, it does not down-regulate NKG2D on NK cells. Soluble form of ULBP2 is frequently detected in sera of patients with hematopoietic malignancies. NKG2D, frequently expressed on tumor cells, stimulate tumor immunity. Soluble forms of ULBPs are also reported to induce protein tyrosine phosphorylation, activation of Janus kinase 2, MAP kinase, and PI-3 kinase/Akt signaling. Clone BUMO-1 specifically detects ULBP2 and is suitable for multiple immunodetection applications. (Ref.: Waldhauer, I., and Steinle, A. (2006). Cancer Res 66(5); 2520-2526; Welte, SA., et al.
