General description

PER2 is a component of the circadian clock mechanism which is essential for generating circadian rhythms. It is a negative element in the circadian transcriptional loop by Influencing clock function by interacting with other circadian regulatory proteins and transporting them to the nucleus. PER2 negatively regulates CLOCK NPAS2-BMAL1 BMAL2-induced transactivation. This gene is a member of the Period family of genes and is expressed in a circadian pattern in the suprachiasmatic nucleus, the primary circadian pacemaker in the mammalian brain. Genes in this family encode components of the circadian rhythms of locomotor activity, metabolism, and behavior. Circadian expression in the suprachiasmatic nucleus continues in constant darkness, and a shift in the light/dark cycle evokes a proportional shift of gene expression in the suprachiasmatic nucleus. The specific function of this gene is not yet known.

Specificity

Cat. # AB2202 will recognize the N-terminus of PER2.

Western Blot Analysis:
C2C12 lysate was resolved by electrophoresis, transferred to PVDF membrane and probed with anti-PER2 (1:500 dilution of this lot). Proteins were visualized using a Donkey anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Quality

Routinely evaluated by western blot on C2C12 lysate.

Target description

Approx. 136 kDa Band may be observed at a much higher 200 kDa by the liver product depending on level of phophorylation and gel system used.

Storage and Stability

Maintain at -20°C in undiluted aliquots for up to 1 year after date of receipt.
