Especificidad

Clone 1N14 is a ZooMAb^® recombinant rabbit monoclonal antibody that specifically detects Telomeric repeat-binding factor 2/TRF2. It targets an epitope within 24 amino acids from the N-terminal region.

Aplicación

Quality Control Testing

Evaluated by Western Blotting in Daudi cell lysate.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected TRF2 in lysates from Daudi cells.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TRF2 in nuclear extract of U-2 OS cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected TRF2 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TRF2 in U-2 OS cells.

Telomeric repeat-binding factor 2 (UniProt: Q15554; also known as TTAGGG repeat-binding factor 2, Telomeric DNA-binding protein, TERF2) is encoded by the TERF2 (also known as TRBF2, TRF2) gene (Gene ID: 7014) in human. TERF2 is homodimeric protein that is ubiquitous in its expression. Higher expression levels are reported in spleen, thymus, prostate, uterus, testis, peripheral blood leukocytes, colon, and small intestine. Two soforms of TERF2 are reported that are produced by alternative splicing. TRF2, like TRF1, contains a Myb-related DNA binding motif. TERF2 binds to telomeric double-stranded 5′-TTAGGG-3′ repeat and plays a central role in telomere maintenance and protection against end-to-end fusion of chromosomes. It is also required to recruit a number of factors and enzymes required for telomere protection, including the sheltering complex, TERF2IP/RAP1 and DCLRE1B/Apollo. TEFR2 colocalizes with telomeric DNA in interphase cells and is located at chromosome ends during metaphase. It is phosphorylated upon DNA damage and the phosphorylated TERF2 is not bound to telomeric DNA, and rapidly localizes to damage sites. Inhibition of TRF2 causes apoptosis in several mammalian cell types and its over-expression is observed in a variety of human cancers. As, TRF2 is a critical component of the telomere protection machinery, with its dysregulation contributing to various pathologies, particularly those related to chromosomal instability and cellular aging. Its roles in telomere maintenance, protection, and length regulation make it a significant focus in the study of aging and cancer biology.This ZooMAb^® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forma física

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitución

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
