Descripción general

JumpStart REDTaq ReadyMix PCR Reaction Mix for High Throughput PCR is formulated with REDTaq Genomic DNA polymerase for amplification of more complex templates and genomic templates. This mix contains optimized enzyme and dye concentrations to provide increased length and yield on the more difficult templates.
JumpStart REDTaq® ReadyMixPCR Reaction Mix is a prepared solution combining the performance benefits of hot start PCR with the dual convenience of Sigma′s ReadyMix and REDTaq. The Master mix includes Sigma′s JumpStart Taq DNA Polymerase, 99% pure deoxynucleotides, buffer, and an inert red dye in a 2× optimized reaction concentrate. Add 25 μL of the ReadyMix to primers, template, and water to a final reaction volume of 50 μL. JumpStart Taq antibody in the reaction mix inactivates the Taq DNA polymerase at room temperature. During the first denaturation step of PCR the complex dissociates and polymerase activity is fully restored. The inert red dye does not effect automated sequencing, restriction enzyme digestion, ligation, or other downstream manipulations. The PCR product is easily separated from the dye by standard purification methods if desired. The hot start mechanism allows for room temperature set up, making it the ideal for high throughput applications.

Aplicación

JumpStartREDTaq®ReadyMix Reaction Mix has been used:
  • For multiplex PCR
  • For amplification with reduced non-specific amplification
  • For high throughput PCR amplification of complex templates
  • In the amplification of primers developed for the pathogenicity-associated genes of Clavibacter michiganensis
  • For PCR amplification of the human TMPRSS6 gene

Características y beneficios

  • JumpStart REDTaq® DNA polymerase, an antibody inactivated hot start enzyme, is designed to minimize non-specific amplification while increasing target yield & specificity
  • JumpStart Taq DNA polymerase provides superior amplification regardless of template concentration
  • REDTaq JumpStart ReadyMix reduces pipetting steps and risk of contamination. The hot start mechanism allows for room temperature set up, making it the ideal for high throughput applications
  • Inert red dye allows for easy verification that reagent has been mixed properly
  • Other loading dyes are not required. Aliquots may be directly loaded onto an agarose gel after the reaction

Artículos

Learn about the history of the polymerase chain reaction (PCR), from the basic principles that proceeded its discovery to the awarding of a Nobel Prize for Chemistry and more recent developments such as real-time PCR (qPCR) and digital PCR.

Real-time polymerase chain reaction allows researchers to estimate the quantity of starting material in a sample. It has a much wider dynamic range of analysis than conventional PCR

Protocolos

Reviews the applications and benefits for RedTaq, including standard RedTaq, Hot Start RedTaq and RedTaq for genomic DNA PCR.

Protocol using antibody mediated hot start polymerase. Method has short activation period (<1min), and results in higher yields and more specificity over standard PCR methods.

When using hot start Taq DNA polymerase, the enzyme remains inactive until heated. Hot Start DNA polymerase control is achieved by chemical or antibody modification of the enzyme.
