Descripción general

GPR18 (G-protein-coupled receptor 18) encodes a seven transmembrane G-protein-linked receptor that is highly expressed in testis and spleen. Its expression is strongest in hypothalamus, thyroid, peripheral blood leucocytes, cerebellum and brain stem. Its expression has been observed to a lesser extent in tissues associated with endocrine and immunologic/hematologic function. The gene is mapped to human chromosome 13q32. The open reading frame (ORF) encoded by this gene spans a length of 993bp. The encoded protein is 331 amino acids long.

Acciones bioquímicas o fisiológicas

The gene GPR18 encodes an orphan G-protein-coupled receptor that binds to N-arachidonylglycine (NAGly), an endogenous lipid that recruits BV-2 microglia and drives cellular migration. GPR18 is activated by phytocannabinoid, Δ9-tetrahydrocannabinol (Δ9-THC), and by NAGly, an oxygenated metabolite of the endogenous cannabinoid anandamide. GPR18 may be involved in central nervous system and endocannabinoid system signaling. It is found to inhibit apoptosis in melanoma metastases and it also facilitates tumor cell survival. It may serve as a potential drug target in metastatic melanoma.
