Descripción general

Gastrin is a pyloric antral hormone, which is a small biologically active peptide thatis synthesized as a precursor. It is found to be produced in antral G cells.

Aplicación

Gastrin I has been used in a study for gastrin stimulation in rats to determine the expression of histidine decarboxylase and histamine metabolism in gastric oxyntic mucosa during conditions of hypergastrinemia and carcinoid tumor. It has also been used for preabsorption of anti-gastrin antibody prior to immunohistochemical assay in rats.

Acciones bioquímicas o fisiológicas

Gastrin plays a crucial role in the maintenance of architecture and function of oxyntic mucosa of stomach, which is involved in the secretion of acid. It also triggers the secretion of acid into the gastric lumen. Continuous increased plasma levels of gastrin or hypergastrinemia, may be involved in gastrointestinal carcinogenesis, especially if it is accompanied with inflammation. The release of this peptide is suppressed during distention, which is a result of omatostatin and intrinsic cholinergic pathways. In rats, this peptide maintains gastric mucosal integrity through the up-regulation of cyclooxygenase-2 which is, in part, mediated by heparin-binding epidermal growth factor-like growth factor (HB-EGF).

Histamine is an important stimulator of gastric acid secretion. In experimental animals, inhibition of acid secretion by long term histamine2 receptor blockade causes hypergastrinemia, proliferation of enterochromaffin-like (ECL) cells, and formation of histamine-producing gastric carcinoids. Gastrin, PGs, and growth factors have important roles in maintaining gastrointestinal mucosal integrity. Cyclooxygenases (COX-1 and COX-2) are the key enzymes involved in PG synthesis. This study aimed to clarify the mechanisms of gastric mucosal protection by gastrin. Fasted rats

We show that the gastric hormone gastrin induces the expression of the prosurvival secretory clusterin (sCLU) in rat adenocarcinoma cells. Clusterin mRNA was still upregulated in the presence of the protein synthesis inhibitor cycloheximide, although at a lower level. This
