ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Especificidad

Clone HM57 is a mouse monoclonal antibody that detects CD79a. It targets an epitope within the C-terminal, cytoplasmic domain.

Inmunógeno

A linear peptide corresponding to 15 amino acids from the C-terminal, cytoplasmic domain of human CD79a.

Aplicación

Quality Control Testing

Evaluated by Flow Cytometry in Human peripheral blood mononuclear cells (PBMC), gated on lymphocytes.

Flow Cytometry Analysis: One million human peripheral blood mononuclear cells (PBMC) lymphocytes were stained with APC conjugated anti-human CD19, fixed, permeabilized and subsequently stained with 5 µL of a 1:1 mixture of Cat. No. CWA-1164, Anti-Human CD79a (HM57) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye. CD79a expression was observed in CD19 positive population.

Tested Applications

No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye. CD79a expression was observed in CD19 positive population.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

B-cell antigen receptor complex-associated protein alpha chain (UniProt: P11912; also known as Ig-alpha, MB-1 membrane glycoprotein, Membrane-bound immunoglobulin-associated protein, Surface IgM-associated protein, CD79a) is encoded by the CD79A (also known as IGA, MB1) gene (Gene ID: 973) in human. CD79a is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-32), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 33-143), a transmembrane domain (aa 144-165), and a cytoplasmic domain (aa 166-226). CD79a is required in cooperation with CD79b for initiation of the signal transduction cascade activated by binding of antigen to the B-cell antigen receptor complex (BCR) that leads to internalization of the complex, trafficking to late endosomes, and antigen presentation. It is also shown to be essential for BCR surface expression and for efficient differentiation of pro- and pre-B-cells. CD79a undergoes phosphorylation on tyrosine, serine, and threonine residues upon B-cell activation. Phosphorylation of tyrosine residues by Src-family kinases is an early and essential feature of the BCR signaling cascade. The phosphorylated tyrosines serve as docking sites for SH2-domain containing kinases, leading to their activation that in turn leads to phosphorylation of downstream targets. Phosphorylation of serine and threonine residues may prevent subsequent tyrosine phosphorylation. CD79a contains an ITAM domain that is localized in amino acids 107-205. Mutations in CD79A gene is known to cause autosomal recessive agammaglobulinemia 3, a primary immunodeficiency characterized by low or absent serum antibodies and low or absent circulating B-cells due to an early block of B-cell development. Forma física

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitución

1.0 mg/mL after reconstitution at 25 µL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 µL PBS. Protect from light.
