Biotin-protein ligase (EC 6.3.4.9; EC 6.3.4.10; EC 6.3.4.11; EC 6.3.4.15; UniProt P50747; also known as Biotin-[acetyl-CoA-carboxylase] ligase, Biotin apo-protein ligase, Biotin-[methylcrotonoyl-CoA-carboxylase] ligase, Biotin-[methylmalonyl-CoA-carboxytransferase] ligase, Biotin-[propionyl-CoA-carboxylase [ATP-hydrolyzing]] ligase, HCS, Holocarboxylase synthetase) is encoded by the HLCS (also known as HCS) gene (Gene ID 3141) in human. HLCS (holocarboxylase synthetase) is the sole biotin protein ligase encoded by the human genome. HLCS catalyses the covalent modification of target lysine residues by biotin. In addiition to taregting various carboxylases in the cytoplasm and mitochondria, both the full-length HLCS and a splicing variant are reported to enter the nucleus and associate with multi-protein gene repression complex. HLCS overexpression in HEK293 cells is reported to downregulate histone H3 lysine9 acetylation (H3K9ac) level with a cocomitant drop in LTRs (long terminal repeats) 15 and 22 due to a decrease in mRNA coding for LTRs. HLCS is also reported to induce RANTES expression in HEK-293 cells by biotinylating heat shock protein 72 (HSP72).
