Alkaline phosphatase (UniProt: P00634; also known as EC:3.1.3.1, APase) is encoded by the phoA (also known as b0383, JW0374) gene (Gene ID: 945041) in E. coli. Alkaline phosphatase is a dimeric, non-glycosylated metalloenzyme, requiring two zinc ions (Zn²⁺) and one magnesium ion (Mg²⁺) per subunit for its activity and structural stability. The active site of Alkaline phosphatase includes a serine residue that plays a crucial role in its catalytic mechanism. The primary function of E. coli Alkaline phosphatase is to catalyze the hydrolysis of phosphate monoesters, resulting in the release of an inorganic phosphate and an alcohol. The enzyme is involved in various processes, including phosphate metabolism and regulation. Alkaline phosphatase is also known for its ability to transfer phosphate groups to acceptor molecules, which is key in its role in transphosphorylation reactions. Alkaline Phosphatase has a wide range of applications in molecular biology. It is used as a reporter enzyme in various assays, including ELISA and Western blotting. During molecular cloning, it is commonly used to dephosphorylate the 5′ ends of DNA fragments to prevent self-ligation during cloning procedures.
