General description

CD40L (cluster of differentiation 40 ligand), a member of TNF (tumor necrosis factor) superfamily, is a membrane bound protein. This protein is predominantly expressed transiently by activated CD4+ T cells either as a cell-surface molecule or a secreted multimer protein. Activated B cells, certain DC (dendritic cell) subsets, platelets, and smooth muscle cells also express this protein.

Biochem/physiol Actions

CD40L (cluster of differentiation 40 ligand) interacts with its receptor CD40 which in turn helps mediate the interaction between APCs (antigen presenting cells) and lymphocytes. Activated platelets produce CD40L which then participates in multiple processes such as endothelial cell reactions, formation of germinal center, T-helper cell priming, and cytotoxic T-cell activation. Platelets are also involved in the pathogenesis of asthma through CD40L where they inhibit the production of regulatory T-cells and promote polarization of T-helper cells. Studies in mice show that this protein confers protection against influenza virus, though in a CD4+ T cell independent manner.
