General description

Immunogen

synthetic peptide corresponding to amino acid residues 1233-1249 of human BACH1 with N-terminal added cysteine, conjugated to KLH.

Application

Anti-BACH1 antibody produced in rabbit has been used in:
  • immunoblotting
  • immunohistochemistry
  • western blotting

Polyclonal anti-BACH1 antibody produced in rabbit is suitable for immunoprecipitation at a concentration of 5-10 μg by using RIPA extract (0.5 mg) of human HeLa cells and for indirect immunofluorescence at 10-20 μg/mL using human MCF-7 cells. The product is also suitable for western blot analysis at a working concentration of 0.5-1.0 μg/mL by using whole extracts of MCF-7 human breast carcinoma cells. Further, anti-BACH1 antibody has shown suitability for microarray analysis.

Biochem/physiol Actions

BACH1 interacts directly with the C-terminal BCRT repeats in Breast cancer type1 protein (BRCA1), a tumor suppressor nuclear phosphoprotein that facilitates the repair of double-stranded DNA breaks. Bach1 also functions as transcription repressor in transfection assays using fibroblast cells and as a transcriptional activator and repressor, respectively, in cultured erythroid cells. Additionally, it is also involved in the coordination of transcription activation and repression by Maf family members such as MafK. Bach1 (BTB and CNC homology 1, basic leucine zipper transcription factor 1) inhibits oxidative stress-inducible genes and is a negative regulator of oxidative stress-induced cellular senescence.
BTB Domain And CNC Homolog 1 (BACH1) interaction between breast cancer type 1 (BRCA1) in vivo appears to be dependent on phosphorylation of Ser990 in the latter during the G2/M phase of the cell cycle. BACH1 mutations were found in patients with early-onset breast cancer. It displays a punctate nuclear staining pattern similar to that observed for BRCA1.
